A group of 200 students from Dinnington Comprehensive will be watching the seven films back-to-back for charity.
Teacher Dana Abdulkarim said the overnight event would be "a test of endurance" for the pupils.
Various students from the school will attend the 'Potter-athon', from first year students through to those who attend sixth form.
The students will gather in the teachers' staff room to watch all the films that have been released so far.
The first film in the series, Harry Potter and the Philosopher's Stone, will be shown on Friday from 1630 BST.
Year 10 student, Charlie Chambers, said "I'm determined to stay awake for the whole marathon. I don't know how I'll stay awake but I will somehow."Fellow student Kaisha Walker said: "I'm nuts about Harry potter, I can't wait, I'm just so excited. We're going to have a really good night."
All of the students attending the event are being sponsored by family and friends to raise money for two causes.
The school which is expecting to raise over ?1,000, will be putting the money towards a school trip to Canada next year and the Sheffield Children's Hospital.
Teachers who have organised the event said there would be minimal breaks during the films.
It is expected that the marathon showing will finish at around 1300 BST on Saturday.
